Chinese scholars published two achievements in the same period

Researchers from the Fudan University Biomedical Research Institute, Peking University Third Hospital, School of Life Sciences and other departments reported the three-dimensional structure of the bone marrow hematopoietic key protein TET2 and new results of in vitro fertilization (IVF) research in Cell magazine.

In the first article, the researchers successfully resolved the three-dimensional structure of the mammalian (mammal; mammal) bone marrow hematopoietic key protein TET2 for the first time. This achievement is of great significance for revealing the pathogenesis of diseases and developing therapeutic drugs for blood tumors (such as myeloid leukemia).

The mammalian TET protein family has three members: TET1 protein, TET2 protein and TET3 protein. If the TET protein loses activity (loss of function), it will cause various diseases, especially blood tumors.

Xu Yanhui's research group carried out research and obtained the three-dimensional structure of TET2 protein and DNA. Using X-ray crystallography and other research methods, the researchers found that the TET2 protein is particularly "smart". It can automatically identify and find methylation markers, and flip the DNA modified with methylation markers into the interior of the TET2 protein. Subsequently, with the help of internal iron ions, the protein undergoes an oxidation reaction, and the label is gradually added to the DNA methylation label.

Further research found that patients with hematological tumors contain multiple TET2 protein mutations, and the mutation rate is very high. The researchers believe that it is precisely because of these mutations that affect the activity of the TET2 protein, causing it to "passively idle" and do nothing, which in turn causes the methylation marker to be "removed", resulting in disease.

In another article, the researchers reported on a safe, accurate, and inexpensive method for selecting genetically normal embryos for IVF procedures, thus increasing the chances of a couple having healthy children.

Researchers have recently developed some whole-genome sequencing methods that can simultaneously detect two defects in a single human sperm cell, but until now, no similar method has been applied to egg cells, and chromosomal abnormalities in egg cells are compared to sperm cells. More common.

In this new study, Professor Suney Xie who worked at Peking University and Harvard University, together with Professor Qiao Jie of Peking University and Researcher Fucho Tang, developed a sequencing polar body Whole genome approach. In the process of female germ cell formation, it undergoes two multiplier divisions to form a large haploid egg cell and 2-3 small cells. These small cells are called polar bodies. Since polar bodies are optional for human embryo development, they can be safely removed without damaging the embryo.
